In vitro study | DAPT functionally inhibits γ-secretase and reduces total Aβ production in HEK 293 cells with an IC50 of 20 nM. DAPT acts on primary cultured human nerve cells and also inhibits Aβ Production. When DAPT acts on all Aβ and Aβ42, IC50 is 115 nM and 200 nM respectively, it is 5-10 times lower than when acting on HEK 293 cells. Recent studies have shown that DAPT inhibits SK-MES-1 cell proliferation in a concentration-dependent manner, with an IC50 of 11.265 μm. In addition, DAPT acts on lung squamous cell carcinoma cells and induces dependent and independent apoptosis by inhibiting Notch receptor signaling pathway. |
In vivo study | DAPT treated PDAPP mice at a dose of 100 mg/kg produced strong and lasting drug effects. The DAPT level in the brain exceeded 100 ng/g within 1 hour and continued to rise for 18 hours after treatment, the highest level of 490 ng/g was observed after 3 hours. During this period, DAPT treatment at A dose of 100 mg/kg also reduced all Aβ and Aβ42 in the cerebral cortex in A dose-dependent manner, with A 50% reduction. DAPT at a dose of 40 mg/kg in the rat cerebral cortex inhibits LPS-induced γ-secretase activity and increases apoptosis in cells carrying long-term neuroinflammation. |
